Readers are book-proportioned hand-held computers with no glare and long battery life which allow you to download and store written material to read (and annotate etc) as you would hard copy. e-Books are digitised works of literature which can be read on a reader. John Siracusa has an engaging piece, The Once and Future E-Book, in Ars Technica which considers, tetchily, the retarded development and inevitably bright future of the e-book - with or without the reader.

PS Don't be put off by the technical references on the first page - this is in fact a very accessible article.
